Absence of viral interference and different susceptibility to interferon between Hepatitis B and C virus in human hepatocyte chimeric mice

چکیده
Aim: Both hepatitis B virus (HBV) and hepatitis C virus (HCV) replicate in the liver and show resistance against innate immunity and interferon (IFN) treatment. Whether 45 there is interference between these two viruses is still controversial. We investigated the interference between these two viruses and the mode of resistance against IFN. Methods: We performed infection experiments of either or both of the two hepatitis viruses in human hepatocyte chimeric mice. Huh7 cell lines with stable production of HBV were also established and transfected with HCV JFH1 clone. Mice and cell lines 50 were treated with IFN. The viral levels in mice sera and culture supernatants and mRNA levels of IFN-stimulated genes were measured. Results: No apparent interference between the two viruses was seen in vivo. Only a small (0.3 log) reduction in serum HBV and a rapid reduction in HCV were observed after IFN treatment irrespective of infection with the other virus. In in vitro studies, no 55 interference between the two viruses was observed. The effect of IFN on each virus was not affected by the presence of the other virus. IFN-induced reductions of viruses in culture supernatants were similar to those in in vivo study. Conclusion: No interference between the two hepatitis viruses exists in the liver in the absence of hepatitis. The mechanisms of IFN resistance of the two viruses target 60 different areas of the IFN system.

Seroprevalence of Hepatitis B, Hepatitis C and Human
Abstract Background Thalassemia patients that are conventionally treated by a regular transfusion regimen are exposed to blood born viral infections. The aim of this study was to investigate the seroprevalence of hepatitis B virus (HBV), hepatitis C virus (HCV) , and human Immunodeficiency virus(HIV) among all multitransfused thalassemia patients in west north of Iran. Material and methods A...
متن کاملDetection of Pre-treatment mutations leading to resistance to direct hepatitis C virus blocking drugs in patients with chronic hepatitis C
Background and objective: Human is the only host of hepatitis C virus. This virus has a positive single stranded RNA and lipoprotein envelop that has 7 confirmed genotypes. According to studies, genotypes 1a, 3a and 1b are the most common genotypes in Iran. No effective vaccine against HCV infection has been developed instead, advances in antiviral treatment using drugs that directly affect spe...
متن کاملPriming Hepatitis B Surface (HBsAg)- and Core Antigen (HBcAg)-Specific Immune Responses by Chimeric, HBcAg with a HBsAg ‘a’ Determinant
We developed an immunogen to stimulate multivalent immunity against hepatitis B surface antigen (HBsAg) and hepatitis B core antigens (HBcAg). Immune responses specific for both HBsAg and HBcAg play an important role in controlling the infection. HBsAg-specific antibodies mediate elimination of virions at an early stage of infection and prevent the spread of virus. The immunogen was constructed...
متن کاملComparison of PEG Interferon Loaded and non-Loaded Iron Oxide Nanoparticles on Hepatitis C Virus Replication in Cell Culture System
Background and Aims: Iron oxide nanoparticles are among the most effective tools which can replace current medical techniques for diagnosis and treatment of various diseases. Hepatitis C infection is one of the main health problems in the world, affecting around 3% of the world's population. This infection can develop into liver cirrhosis and liver cancer over the time in 80% of patients. In t...
متن کاملIntrinsic Viral Factors Are the Dominant Determinants of the Hepatitis C Virus Response to Interferon Alpha Treatment in Chimeric Mice
BACKGROUND Hepatitis C virus infection is a global health problem. New direct-acting antiviral agents have been recently approved. However, due to their high cost and some genotypes remaining difficult to treat, interferon-based therapy with pegylated interferon and ribavirin likely may remain a component of hepatitis C virus treatment for some patients. Unfortunately, pegylated interferon / ri...
